Streaked out B. subtilis 168 & B. subtilis ComI on LB agar from -80°C stock on LB agar plates and grown overnight at 37 °C.

Picked up a colony from both 168 & ComI plate strain and put in LB medium (3 mL in culture tube). Grown overnight in shaker incubator (37 °C, 220 rpm).

168 DNA (50 µL, 21.9 ng/mL DNA)
