A colony PCR was done to check if the ligation of the PGA into pSB1C3 was successful.
Make a more negatively charged biofilm.
The insert was not the correct size.

Transformation showed colonies. 8 colonies were tested using a colony PCR. All colonies did not show the right band. So all plasmids did not contained the right insert.
