IGEM-SAFETY

Does your project include any security features? Do you make any design to reduce risk?

Choose one non-pathogenic chassis

1. Carrier choice: We choose the carrier (pSB1K3 pSB1C3)which is recommended and provided by IGEM and use no other source of the carrier, to prevent the spread of the virus and the spread of antibiotic resistance genes in the laboratory and in nature

2. The choice of the host: We use the most commonly used, the most worldwide, the maturest and the securest systems. The host DH5 alpha and express host bacterium BL21are safe to operators, the laboratory environment and the external natural environment, which are recognized throughout the world.

3. No harm to human/animal/plant

We only use E.coli, which is commonly used in the lab, relatively friendly to the environment and does not infect plants. E. coli is a intestinal endophyte in human and animals, existing in human's and animal's body and will not cause disease to human and animal.

4. Safer material instead of dangerous materials/ testified experiments

Host bacterium DH5 alpha and BL21 for laboratory and biological pharmaceutical industry are the most commonly used host bacteria systems. Compared with the wild E. coli in the nature, they are more controllable, less harmful and better safer.

5. Include a "induced killer" or the "lock" equipment

In order to enhance security, we eliminate the possible damage to the operator and the outside environment. This project contains a light-induced biological suicide program. After finishing the pesticide degradation process, the bacteria can accept uv-irradiation, activate the promoter of RecA and the promoter of ccdB suicide gene expression which produce ccdB protein. This protein can inactivate the host DNA gyrase, resulting in the death of the host cell to eliminate noxious impact to the environment.

What security strategy do you use every day in the laboratory? Have you done any unusual experiment, or face any unusual security problem?

1. All operations are limited in the laboratory environment. Experiments which involve bacteria parts are completed in a super clean workbench. All operations accord to the regulations of the laboratory safety instructions. Operators wear disposable gloves, hats, masks and safety protection equipment.

2. All organic solvent experiments are completed in the fume hood, and operators wear protective masks to avoid the harm of organic solvent steam. 3.Waste classification and sterilization accord to the regulations of the laboratory safety. All bacteria and DNA experiment equipments are disinfected by high pressure steam before discarding.

Do you face any safety problems when sending your DNA part? How do you solve these problems?

To submit the corresponding biological module, we do all procedure in accordance with the requirements of IGEM. The solid form plasmid is sealed in porous plate, according to the requirements of the igem seal plate with film sealed preservation, to avoid the leakage and diffusion in the process of transportation.
