IGEM-SAFETY

1.Pick up one non-pathogenic chassis

Selection of carrier: pick up the carrier(pSB1C3)recommended and provided by igem. Carriers from any other sources are not applicable, which avoids the spread of viral vectors and antibiotics resistance genes in the lab and nature.

Selection of host strain: Operate the world’s most commonly used, maturest and safest system in the lab. Use DH5αto clone host strain and adopt BL21 to express host strain. The safety of operating personnel, lab and natural environment is universally acknowledged all over the world.

2.Selection parts, no harm to human beings,animals,plants

The escherichia colitic, frequently used in the lab, established in the biology module as genetic engineering bacteria is friendly to environment and doesn’t infect plants. As the endophyte in the intestinal tract, it lives inside human beings and animals. It won’t produce any harm to human beings and animals under natural conditions.

3.Safer material replaces dangerous one.

The chosen DH5α and BL21, as the most used host strains system in labs and biopharmaceutical industry have the feature of better artificial controllability, much safer and lower pathogenecity, compared with escherichia colitic in the wild.

4.Include the equipment with " inducing lethality" or "refusal of entrance"

To further strengthen security and remove any chance of harm to operating personnel and external environment, the project is designed with biology module of enzy-matiCphotoreactivation inducing suicide programme. When finishing degradation of pesticide, genetic engineering bacteria activates promoter in the repair enzyme of biology module after receiving UV. The promoter launches the expression of downstream suicide gene and produces CCDB protein which can inactivate DNA gyrase of host strain and lead the death of host strain cell. Thus, the influence of genetic engineering bacteria to the outside environment would be wiped out.

All operations should be conducted under limited conditions. Any lab sections related to bacteria are required to be completed on the clean bench. Lab staff must wear lab coat, disposable gloves, cap, respirator and other safety protective products according to lab regulations.

Any lab sections related to organic solvent are required to be completed in the fume hood. Wearing protective head-cover to avoid steam of organic solvent to cause any harm to operator or other lab staff. Test waste should be processed by their classification on the basis of lab regulations. All lab appliances contaminated by bacteria and DNA must go through bacteria disinfection by high pressure steam before discarding which then are treated with classification disposal.

The delivery of relevant biology modules to igem is based on the normalized requirements of igem. The plasmid, kept as solid form, is sealed off in the perforated plate and capped with microplate sealer to avoid leak and spread during transport.
