tasA PliaG Heat shock transformation (t72)
The ligation product of tasA and PliaG was transformed into E. coli using a heat shock transformation.
Our goal is to overexpress TasA by adding a second copy of the tasA gene under the control of a salt inducible promoter. This overexpression will lead to a stronger biofilm when the biofilm is exposed to salt water.
The transformation of tasA with PliaG in E. coli was successful.

A heat shock transformation was performed with competent cells. The culture was plated on LB+cm (5ng/mL). The plate was put 5h at 37 °C, 48h at room temperature and 4h at 37 °C.
