Design
T25
The constructs LacZ promotor, is induced by IPTG and inhibited by LacI and CAP. It’s followed by a ribosomal binding site. The coding region is KT25 subunit of adenylate cyclase, followed by a linker, the wanted target protein and a terminator. T25 origins from Bordetella pertussis. Backbone for the construct is pSB1K3.
T18
The same promoter regulates this construct as with T25. After the promoter, it contains a ribosomal binding site, the other subunit of Bordetella pertussis adenylate cyclase T18, a linker, the scaffold human thioredoxin containing a XhoI site, and a 3xFLAG tag. pSB1C3 is this constructs backbone.
