Use the protocol of the Sigma-Aldrich GenElute™ Bacterial Genomic DNA Kitto purify Gram positive bacteria DNA out of agarose gel. Instead of adding binding buffer in a 1:1 ratio to the DNA sample as noted in the protocol, add binding buffer a 2:1 ratio to the DNA sample. To dissolve the gel in the binding buffer, vortex the sample.
