PCR of the ste2 gene from the genome of Saccharomyces cerevisiae 7284 strain.
- Genome extraction
- PCR - annealing temperature was 55°C, 30 cycles
- Gel electrophoresis - verification of successfull amplification of ste2

- Restriction digest - restriction of the amplified ste2 gene and pRSII416 vector with BamHI + HindIII
Tuesday
- Gel electrophoresis - pRSII416 from this Monday
- Gel purification - extraction and purification of the digested pRSII416 vector
- Ligation - 26.4ng of the insert
- Transformation - 5μL of the ligation was transformed into E5alpha cells. After 2h of incubation at 37°C, 300μL was spread on Amp plate.
