To 5 ul from each plasmid was added: 13 ul MiliQ 1 ul fast digest EcoRV 1 ul fast digest buffer incubated for 1 hour at 37C Good plasmid would have 2 bands K823023 (original) or pdg1664 (original) would have 1 band. All 5 plasmids had two bands. One of the plasmids will be used further. named as thrC plasmid A
