Transformation into not competent E.coli cells
- grow overnight culture
- Dilute1:100
- grow 10 mL day culture 3h @ 37 °C
- take 1 mL
- centrifuge (1min, 4000 rpm, 4 °C)
- resuspend the pellet in 100 µL of TSS
- add 1 µL of plasmid (~200 ng)
- incubate 30-60 min on ice
- heatshock @ 42 °C for 1 min
- incubate 15 min on ice
- add 1 mL LB-broth
- incubate 1h on thermoshaker (37 °C, 400 rpm)
- centrifuge (10min, 4000 rpm)
- resuspend in 50 µL residual media
- plate on plate
- incubate @ 37 °C over night
