Crystal Violet Staining

Preparation

• grow cells in 96 well plate
• use TB-medium
• incubate for 24 h / 48 h

Washing and staining

• remove liquid
• wash wells with sterile water (3 times)
• add 150 µl crystal violet solution (1%, in water)
• incubate for 20 min
• remove crystal violet solution
• wash wells until liquid is uncoloured
• dry wells

Plate reader measurement

• add 200 µl ethanol (98%)
• incubate for 1 h, while shaking the plate (the crystal violet has to be dissolved)
• measure absorbance at 595 nm
