Competent E. coli cells (RbCl)
- inoculate 100 mL of LB-Media with E. coli and let the cells grown to OD600 = 0.5
- incubate the culture on ice for 10 minutes
- centrifuge the culture (2 x 50 mL, 10 min, 3000 rpm, 4 °C)
- discard the supernatant
- wash the pellet with 10 mL TFB1
- centrifuge (10 min, 3000 rpm, 4 °C)
- discard the supernatant
- resuspend the pellet in 2 mL TFB2
- pipette 200 µL aliquots
- cool it in liquid nitrogen
- stored @ -80 °C
